| Research Abstract |
1. 扁平上皮癌に関して
(1) ヒト食道扁平上皮癌の転移巣から得られた培養株より分離・樹立したトロンボモジュリン(TM)の高発現株と低発現株を比較して、低発現株の方がより高い浸潤性を示すことを明らかし、また、TMが扁平上皮癌の血管新生能、及び接着能に関与することも見い出した。
(2) ヒトの口腔や肺の扁平上皮癌において、食道癌と同様にリンパ節転移巣でTMの発現低下が起こることを明らかにした。
(3) ヒト食道癌において、シアル酸の結合したMUC1ムチンの陽性群は陰性群に較べて明らかに予後が悪かったが、MUC1ムチンコア蛋白の陽性群と陰性群の間には予後の差はなかった。
2. 腺癌に関して
(1) 膵と肝内胆管の膨脹性発育腫瘍でも、その経過のうちに浸潤性増殖を始める症例においては、浸潤性発育部位で、予後不良因子の膜結合型MUC1ムチンが発現し、腫瘍の浸潤にかかわっている可能性が見い出された。
(2) 浸潤傾向の少ない「粘液産生性膵腫瘍」の各組織亜型でのムチン発現の検索により、分泌型ムチンのMUC5AC(胃型)とMUC2(腸型)の遺伝子発現パターンが各組織亜型により異なっていることを明らかにした。
(3) 膵と肝内胆管の浸潤性癌と膨張性癌においてシアリダーゼ処理によるコア蛋白と糖鎖の結合状況を検討する研究を行い、浸潤性癌に特徴的なMUC1ムチンにおいてはシアル酸エピトープが重要であることを明らかにした。
(4) ヒト胃癌において、糖鎖の結合状況の異なる種々のMUC1ムチンと、MUC2ムチンの発現を検索したところ、MUC1ムチンの糖鎖の結合状況に関係なく、MUC1(+)・MUC2(-)のグループが最も予後が悪く、MUC1(-)・MUC2(+)のグループが最も予後が良いことが明らかとなった。
(5) 大腸でのadenoma-carcinoma sequenceにおける腫瘍の進展とpericryptal fibroblast sheathの減少に密接な関連性があることを明らかにした。
