Research Abstract	IL-12はT細胞機能を著明に亢進して全身性または組織局所的炎症を惹起させ、移植片拒絶反応、自己免疫等炎症患の成立及びその慢性化に深くかかわっている。その為IL-12の作用を制御することが各種病態の改善に連がることが考えられる。そこで本年度の研究はIL-12産生の細胞性・分子機構を解析することを目的とし、次の成果を得た。 IL-12は主にマクロファージ(MΦ)/antigen-presenting cells(APC)より産生されることがわかっている。しかしその産生様式は単一ではない。即ち、結核菌、LPSなどの病原微生物又はその関連物質でIL-12が惹起される他に、T細胞-APC相互作用によっても誘導されることがわかっている。我々もいくつかのTh1タイプクローンとAPCのinteraction、及びT細胞レセプター(TCR)刺激を受けたnaive T細胞とAPCのinteractionでIL-12が産生されることを見い出した。そこでいかなる分子間interactionがIL-12産生を惹起するかについて解析し、次の結果を得た。即ち、APCより抗原刺激を受けたT細胞は、数時間内に一過性にCD40 ligand(CD40L)を発現した。fresh T細胞の場合はCD4^+画分のT細胞のみCD40Lを発現した。更に、CD40LはAPCのCD40を刺激し、APCによるIL-12産生が誘導されることが、抗原刺激を受けているT-APC相互作用中に抗CD40L抗体を添加する又はCD40欠損マウスよりのAPCを用いるとIL-12産生が誘導されないことにより証明された。
