| Research Abstract |
抗リン脂質抗体(抗カルジオリピン抗体;aCLおよびループスアンチコアグラント;LA)の存在は、血栓症、子宮内胎児死亡および血小板減少症と関係がある。このような病態にたいして、抗リン脂質抗体症候群の名称がつけられているが、本症候群の臨床像は抗リン脂質抗体に起因する血栓形成である。本研究では抗リン脂質抗体、特に抗カルジオリピン抗体の詳細な特異性を解析し、以下の点を明らかにした。
1)酸素原子を導入したポリスチレンプレートや、カルジオリピンやホスファチジルセリンなどの陰性荷電リン脂質とβ2-GPIとの接触により、β2-GPIの抗原構造に変化が生じ、抗リン脂質抗体症候群患者由来の抗カルジオリピン抗体がβ2-GPIを認識するようになる。
2)上記の実験結果に基づき、カルジオリピンを抗原として用いない、新しいaCLの測定方法を確立し臨床的有用性を明らかにした。
3)in vitroにおけるβ2-GPIの生物活性を、β2-GPIの欠失変異蛋白および領域特異的なモノクローナル抗β2-GPI抗体を用いて解析した。
4)β-GPIが酸化LDLに結合して、マクロファージによる酸化LDLの取り込みを抑制する事を発見した。逆に、酸化LDLへの結合は、ヒトβ2-GPIとモノクローナルaCLの同時添加により、有意に増強した。
