Research Abstract |
The cause or the etiopathogenesis of rheumatic diseases including rheumatoid arthritis is unknown. It is however thought to be caused by both environment factors and genetic predisposition. As to the genetic predisposition, polygenic origin is hypothesized. Nevertheless, the detail of the genetic predisposition is almost totally unknown. The present study was to identify the disease-susceptible gene loci on the chromosomes for rheumatoid arthritis. METHODS DNA was extracted by using guanidium salt technique from two sib-pair of rheumatoid arthritis and one healthy sibling. Microsatellite markers at 385 points on total 22 chromosomes in a distance of approximately 7.6 cM (mean) was amplified by using fluorescent-tagged DNA primers. Amplified products were electrophoresed in a ABI377 DNA sequencer and amplified marker DNAs were sized. Inheritance of each of the sized DNA was analyzed in each sibling family. We have analyzed 35 different rheumatoid families. The result was now being analyzed and the result so far indicated that chromosome markers D2s165, D3s1565, D5s406, D5s424, D7s515, D8s556, D9s158, D11s898, D12s99 and D21s263 were significant and showed a Lod score exceeding 1. We identify the disease susceptible loci from this basic data.
