| Research Abstract |
本研究計画全体は牧野によって統括され、2プロジェクトが並行して順調に進行中である。平成9年度の成果は以下の様にまとめられる。
1)主要組織適合抗原クラスI(MHC class I)遺伝子発現調節機構能の解析
ヒト株化培養肝細胞において、
(i)胆汁酸で活性化されるプロテインキナーゼCアイソフォームを推定した。
(ii)胆汁酸によってMHC class I遺伝子プロモーターの誘導が起こることを明らかにした。
2)ウルソデオキシコール酸の免疫調節作用機構の解明
ウルソデオキシコール酸によるステロイドホルモン受容体の翻訳後修飾機構の一部を解明した。
Geldanamycin,herbimycin Aを用いた実験から、ウルソデオキシコール酸は細胞質において、熱ショックと結合している安定型ステロイドホルモン受容体とは結合せず、非結合型と結合して核に移行させる可能性が高いことが判明した。本発見は世界的にも画期的であり、今後のステロイドホルモン受容体を標的とした創薬に多大な影響を与える可能性がある。
