Research Abstract

1996年、CCK-Releaing-Peptide(lumina CCK releasing factor=LCRF)が、部分精製された。このペプチドは、分子量が約8000であるが41個しかまだあきらかになっていない。我々は、1-35のfragmentを合成し、覚醒ラットにおいて、それが生物活性をもつことを確認した。さらにペプチドのfull-lengthを確定するため、抗体を作成した。この抗体を用いて、小腸内腔の濃度を測定した。また免疫組織学的に小腸粘膜を染色したところ、小腸粘膜絨毛先端が強く染色された。これからaffinity columnを作成して、CCK-RPの精製を行う。
CCK-Aレセプター欠損ラットにおける病態生理の検討として、本系のラットの遺伝子の異常部位を確認した。CCK-Aレセプター遺伝子は約10kbからなり、5つのexonと4つのintronからなる。本系のラットでは、プロモーター領域からexon2にかけて、大きな欠損がみられた。膵内外分泌機能は、CCKに対して全く反応しなかったが、neuromedin,secretinなど他の刺激に対しては正常ラットと同様の反応を示した。身体の成長に伴う膵の成長の変化は、哺乳期にはの正常ラットと同様であるが、離乳開始より、膵重量の増加が遅延した。CCK-AレセプターmRNAは、胎児期には正常ラットでも発現せず、CCK-BレセプターmRNAのみ発現がみられた。生後CCK-AレセプターmRNAは増加し、逆にCCK-BレセプターmRNAは減少して、離乳期には検出されなくなった。膵臓部分摘出後の膵臓の再生はCCK-Aレセプター欠損ラットでは、正常ラットに比較し、遅延していた。実験的に膵障害(膵管結紮による)をおこした場合、急性障害度は正常ラットと同様であったが、再生が遅延していた。
