Research Abstract

1。1996年、luminal CCK releasing factor(LCRF)が部分精製された。N-末端より41個のアミノ酸が同定されたが、全残基はまだ不明である。我々は、LCRF1-35を合成し、抗体を作成、RIAをestablishし、LCRFの全アミノ酸構造決定を試みるとともにLCRF1-35の生物活性を確認した。さらに、分泌動態を検討している。
2。ラット、マウス、ヒトのCCK-A受容体遺伝子のゲノムクローニングを完了した。ラットのCCD-Aレセプター遺伝子は、14番染色体にあり、約10kb,5exon,4intronからなる。肥満ラットは、プロモーター領域を含む約6.8kbにわたって欠損がみられた。
3。膵臓部分摘出後の膵臓の再生はCCK-Aレセプター欠損ラットでは、正常ラットに比較し、遅延していた。実験的に膵障害(膵管結紮による)をおこした場合、急性障害度は正常ラットと同様であったが、再生が遅延していた。胃において、CCK-Aレセプター欠損ラットでは、CCK-B/gastrinレセプターの遺伝子発現が増加しており、胃酸分泌が亢進、壁細胞の肥大がみられた。CCK分泌はCCK-Aレセプターの存否には影響されなかった。
4。CCK-Aレセプター欠損ラットでは、光に対する視索上核の反応が障害されていた。
5。拘束ストレスに対する、リンパ球のsubpopulationは、CCK-Aレセプター欠損と正常ラットとの間に差がみられなかった。
6。CCKは、胃の迷走神経求心路を興奮させるが、これはCCK-Aレセプターを介して生じることを確認した。
7。CCK-Aレセプター欠損ラットでは、小腸内刷子縁膜酵素活性が増加していた。
8。CCK-A受容体遺伝子ノックアウトマウスを作成した。
