炎症性・増殖性皮膚疾患における各種デスモソーム構成蛋白の関与の分子生物学的検討

Research Abstract	昨年に引き続き、デスモヨ-キン(DY)の細胞膜および核内への移行に関与する分子内部位を明らかにする目的で、そのN末端・中央部・C末端部のcDNAを組み込んだ有核細胞発現系ベクター(pcDNAI/Amp)のconstractを用い、今回はMDCK細胞および正常ケラチノサイトにtransfectした。その結果、DYのN末端および中央部は細胞質内に分布し細胞膜および核内への移行は見られなかったが、C末端部には核内および細胞膜部両方への移行が見られた。これはDYのC末端部が細胞接着と細胞内伝達の両方に重要な機能を有していることを示したものといえる。さらに、私共は、各種の免疫学的・生化学的手法を用いた検索で、ごく最近新しく発見されたデスモソーム蛋白で、cornified envelopeとの結合に関与していると考えられている、エンボプラキンおよびペリプラキンが、新しい疾患概念である腫瘍随伴性天疱瘡の抗原であることを証明した。これらの蛋白は表皮ケラチノサイトの細胞接着および分化に重要な役割を果たしていると考えられる。今後、尋常性乾癬、有棘細胞癌、扁平苔癬などの多くの炎症性・増殖性皮膚疾患の発症における関与の可能性について検討する予定である。一方、デスモソーム細胞接着蛋白としては、各種デスモグレイン(Dsg)が様々な水疱症の抗原となりうること、特に腫瘍随伴性天疱瘡の抗原のひとつである可能性を示唆した。さらに、もう一種のデスモソーム細胞接着蛋白であるデスモコリン(Dsc)の皮膚疾患における重要性はいまだ報告がなかったが、今回、Dsc1が新しい疾患概念であるIgA天疱瘡(SPD型)の自己抗原であることを証明した。これは、この疾患には著明な膿疱形成が見られることより、Dscが細胞接着だけではなく好中球の遊走など各種炎症にも関与していることを示唆した。
