| Research Abstract |
当初の予定に準じ研究を施行、これまでに以下の結果を得ている。
1.担癌宿主末梢組織におけるインスリン抵抗性介在の有無 これまでに約40例の患者6例のvolunteerでインスリン抵抗性存在の有無をeuglycemic hyperinsulinemic glucose clampをもちいて検討、担癌患者では、高頻度にinsulin resistanceが存在すること、これらはstage、原発巣の部位〔胃、大腸,肺〕によらないこと、細胞内糖代謝の変動からみた意味は著明はglucose torageの減少と有意なglucose oxidationの反映であることを見いだした(現在3編の論文を準備または投稿中である)。
2.末梢組織における組織内glycogenの測定 組織内glycogen量測定方法を樹立、末梢筋肉内glycogen量がindirect calorimetry及びglucose calmpから得られたM値、glucose storageと有意に相関することを見い出した。(7月学会発表予定)。
3.glucose transporterの解明 ひと胃癌細胞(MKN28、STSA、MKN45など)を用い、株によりGlut1-5の発現に差があること(Immunohistochemistry)、Glut4発現の有無によりglucose transportに有意な差があること(glucose transport assay)(3月のSociety of Surgical Oncology、Chicagoに採用、発表予定)、更に現在施行中の研究で癌細胞のGlut4 mRNAには特異な変異がしょうじていることが強く疑われる結果を得ている(rt-PCRによる検討)〔本年度の癌学会及び来年度のAmerican Association for Cancer Researchに投稿予定〕。
平成9年度 骨格筋内Glut4mRNAと癌細胞内Glut4mRNAとの間に有意な差異がありそうな結果となってきており、現在いくつかのprimerを作り替え詳細な検討にはいっている。これらを更に掘り下げると共に、apoptosis、telomeraseとの関連を明らかにする。これら方法論の方法論は8年度に試しておりほぼ信頼できる結果が得られるようになりつつある。
