Research Abstract |
Contact-inhibited fibroblast cell lines have been established from the homozygous C3H/Cs^b mutant mouse embryo and wild type C3H/Cs^a mouse embryo. The catalase enzyme activities of the wild -type C3H/Cs^a cells and mutant cells were 10.4 and 2.3 U/mg protein at passage 29, respectively. However, there was no difference in the activity of glutathione peroxidase and superoxide dismutase between two cell lines. As to catalase, it is interesting that catalase activity of wild-type C3H/Cs^a cells decreased with in vitro culture aging. The catalase-deficient C3H/Cs^b cells were markedly sensitive to the hydrogen peroxide, compared to wild-type C3H/Cs^a cells. Furthermore, the cells were sensitive to X-rays and Near-UV.However, there was no difference in the sensitivity of the topoisomerase II inhibitor, mAMSA, and the cross-linking agent, cis Pt, between two cell lines. 8-Hydoroxyguanine is one of the major modified dases in DNA induced by reactive oxygen species. 8-Hydoroxyguanine content per 10^5 dG was observed three time increase in mutant cell lines than that in wild cell lines at passage 27. These cell lines will be of use in the study of the role that catalase plays in the intracellular prevention of DNA damages induced by oxidative stress.
