| Research Abstract |
(1)C3植物(イネ)よりC4光合成酵素の一つであるピルビン酸Piジキナーゼ(PPDK)遺伝子を単離し、そのプロモーター領域をクローン化した。その塩基配列をC4植物(トウモロコシ)のPPDK遺伝子のそれと比較したところ、類似した領域とそうではない領域が存在していることがわかった。
(2)トウモロコシPPDK遺伝子のプロモーター配列を5'末端から削除したプロモーター断片を準備し、これらのプロモーター断片とGUS遺伝子を結合したキメラ遺伝子を作成した。これらのキメラ遺伝子をトウモロコシの葉肉細胞にエレクトロポレーションにより導入してGUS活性を測定することによりプロモーターの活性の強さを測定したところ、トウモロコシPPDK遺伝子の高いレベルの発現には-174から-83までの領域が重要であることがわかった。
