Research Abstract

A membrane enzyme needs a lipid surrounding for the activation. Therefore, the immobilizing carrier must have a lipid surrounding on itself when a membrane enzyme is an immobilized to a carrier. In this work, a giant lipid-vesicle of hybird type (5-50mum in diameter) was tried to apply as an immobilizing carrier of a membrane enzyme and following results were obtained.
(1) Fluctose dehydorgenase (FDH) and some other kinds of bioactive agents could be also immobilized by means of a second-stage emulsification.
(2) By means of a membrane filtration (pore size = 5mum), the enzyme-immobilized vesicle could be successfully recovered and the specific activity lowering of the immobilized FDH was found to be only 5- 8% per a operation of the immobilized-FDH reaction.
(3) By incoporating either BSA or gamma-globulin into a lipid membrane of a carrier vesicle, the activity of the immobilized FDH to the vesicle, which coexisted with the incorporated protein, was enhanced in the range of certain concentrations of the proteins. The FDH reaction was found to be entropy-dirven in the case of BSA as an coexisting protein and enthalpy driven in the case of gamma-globulin.
(4) The specific activity of an immobilized FDH linearly correspounded to both the helix content of the immobilized FDH and the fluidity of the lipid membrane of the carrier vesicle.
Thus, the vesicle was found to function to activate the immobilized membrane enzyme.
