消化管癌における細胞周期調節因子の遺伝子異常と遺伝子・染色体不安定性との関係

Research Abstract

1. 消化管癌における遺伝子および染色体の不安定性
胃癌生検組織および手術材料について、D1S191,D17S855,BATRII,BAT40の4つのローカスの遺伝子不安定性をマイクロサテライト法で検討した。4領域中2領域以上でreplication error(RER)をしめす症例(mutator phenotype)は、胃癌全体の約3%であったが、これらの50%では、胃あるいは他臓器に別の癌が存在していた(多発癌)。さらに、一部ではミスマッチ修復系遺伝子の生殖細胞異変が認められた。
胃癌および大腸癌について、テロメラーゼ活性および各種テロメラーゼコンポーネントの発現を検索した。胃癌、大腸癌どちらにおいても、大部分でテロメラーゼの強い活性と触媒サブユニットTERTの過剰発現が認められ、両者はよく相関していた。胃の前癌性病変である腺種においても、低いレベルながらテロメラーゼ活性が存在し、また腺種および腸上皮化生の一部において、TERTの発現が見いだされた。
2. 消化管癌における細胞周期調節因子の遺伝子異常
胃癌、大腸癌について、細胞周期調節因子の標的である転写因子E2Fの遺伝子異常および発現を検討した。E2F-1遺伝子増幅は、胃癌、大腸癌でそれぞれ4%、25%に認められ、胃癌の40%において過剰発現が認められた。一方、E2F-3の発現は、70%の胃癌症例において減弱していた。
CDKインヒビターp27の発現を胃癌原発巣と転移巣で比較すると、原発巣においてp27の発現が不均一で、転移巣での発現が殆どみられないものが多く、cyclinEの発現は、原発巣と転移巣で差は見られなかった。
