NK細胞と主要組織適合遺伝子複合体ClassI分子の関連

Research Abstract

1)現在ヒトのNK細胞より抗CD3、抗CD4、抗CD8、抗CD19抗体を利用しNK細胞をnegative selectionにより精製したが、NK細胞の長期維持には失敗している。精製されたNK細胞をNK細胞receptorに対する抗体であるEB6(Cw2,4,5,6)、GL183(Cw1,3,7,8)、DX9(Bw4)を利用しHLAの異なる患者において検討しているが、一定の傾向を認めていない。すなわちCw2,4,5,6を有しないヒトでもEB6陽性のNK細胞を認め、逆にCw1,3,7,8を有しないヒトにもGL183を認める。現在、NK細胞cloneからの抗体作製をあきらめ、精製NK細胞をからの新しいNK細胞receptorに対する抗体の作製中である。上記の方法でNK細胞(A2402/A2402,/B0702/B0702,Cw0702/Cw0702)を精製し、Balb/cmiceに免役し、NK細胞株NK-92(A0301/A1101,B0702/B44031,Cw0702/Cw1601)を利用しflow cytometryでscreeningを実施し、現在17cloneを分離している。更にNK-92の感受性株(K562)あるいは耐性株(SBC-9A)を利用し,function assayを実施しinhibitoryあるいはactivate receptorの分離同定中である。
2)NK細胞においてNegative signalに関与するNK細胞receptorの一つであるLy-49Aのcytoplasmic domain(ITIM like motif,VxYxxVを持つ)とCD3ζのtransmebraneとCD16のouter membraneのchimeraを作製し、これをB細胞であるA20にtansfectionした。このtransfectionされた細胞では抗CD16抗体とヤギ抗マウスIgGにより部分的にIL-2の産生が抑制され、B細胞において確立されているITIMmotifによるnegative signalと類似のsignalがNK細胞においても働いていることが推定されたが、これにはCa++は関与しておらずこの機構ついて現在解析中である。
