Research Abstract	本年度はRNAポリメラーゼII(RNAP II)に対する自己抗体の分子免疫学的性状,免疫遺学的背景を追究した。【方法】(1)免疫沈降法でRNAP II subunitと反応した抗RNAP II抗体陽性の強皮症15例を対象,全身性エリテマトーデス、多発性筋炎,健常人の589例を対照とした。(2)免疫沈降反応により,RNAP II large subunitの220kDa脱リン酸化型,240kDaリン酸化型subunitとの反応性を調べた。さらに,18アミノ酸からなるCTD合成ペプチド{NH_2-(YSPTSPS)_2-YSPT-amide}を添加し,かかる反応に対する抑制活性も検討した。(3)pulse chase analysisにより同抗体の経時的RNAP II subunitの認識様式を検討した。(4)抗RNAP抗体陽性例の末梢血白血球より抽出したgenomic DNAを用い,HLA Class II(DRB1,DQA1,DQB1)遺伝子をPCR-RFLP法で調べた。【結果】(1)抗RNAP II抗体陽性15例は全例220kDa脱リン酸化型,240kDaリン酸化型の両subunitを認識した。合成ペプチドによる免疫沈降反応抑制試験では,11例中8例(5例で完全,3例で部分的)に抑制が認められた。かかる例では,両subunitに対する反応が抑制された。(2)抗RNAP II mAbである8WG16および抗RNAP II抗体陽性血清は先ず220kDa脱リン酸化型subunitのみを認識,経時的に240kDaリン酸化型subunitを認識するようになった。これは220kDa subunitのposttranslational modification過程に一致するものと考えられた。さらに,もう一つのlarge subunitである145kDa subunit蛋白との反応性が経時的に増加した。(3)抗RNAP抗体陽性例のHLA Class IIの検討では,DR2(DRB1^1502あるいはDRB1^1501)を83%に認め,健常人コントロールに比し,有意に高頻度であった(p<0.05)。【結語】RNAP II-220kDa subunitのリン酸化型、脱リン酸化型の両subunit CTDが抗RNAP II抗体の主要エピトープと考えられた。また、同抗体産生とHLA Class II遺伝子との関連が示唆された。今後,症例を集積しより詳細な分析を行う予定である。
