Research Abstract |
We investiqated the potential role of a variety of growth factors in the regulation of mucosal repair and remodelling processes in the airways of asthma. Among growth factors ppssibly accumulated in the local environments of the airways, transforming growth factor-beta-1(TGFbeta-1)showed a potent inhibitory effect on the proliferation of human bronchial epithelial cells. A Th1-T cell-derived lymphokine interferon gamma also showed an inhibitory action on the cell growth. Moreover, TGFbeta-1, but not interferon-gamma, had a suppressive effect on the ciliated differentiation of human airway epithelial cells as assessed by an in vitro model of air-liquid interface culture. The megnitude of TGFbeta-1 gene expression was uprehulated in the qpithelium obtained from patients with airway inflammatory disorders including asthma as evaluated by a semiquantitative reverse transcription and polymerase reaction technique. TGFbeta-1 has been reported to stimulate airway fibroblasts to produce collagens and extracellular matrices. Taken together, therefore, it may play an important role in the repair and remodelling responses of the airways.
