生理活性ペプチドによる血管のオートクリン-パラクリン型緊張・増殖制御機構の解明

Research Abstract	本研究の目的は生理活性ペプチドによるオートクリン作用について明らかにすることである。まず、アドレノメジュリンが、どの様な細胞に発現し、どの様な細胞に作用を及ぼすかについて明らかにした。その結果、アドレノメジュリンが心臓、腸管、血管、膀胱、気道等のいろいろな細胞に発現していることが明らかとなった。しかしながら、その平滑筋に対する作用は、限られていることがわかった(Nishimura et al.,Br.J.Pharmacol,1997)。また、bradykinin(Eguchi et al.,Br.J.Pharmacol,1997)等の生理活性ペプチドに対する血管作用を明らかにした。気道におけるendothelinのオートクリン作用について検討した結果、気道平滑筋、気道上皮にpreproendothelin-1,preproendothelin-3,endothelin receptor,endothelin converting enzymeが発現しており、オートクリン作用の存在が明らかとなった(Yoshimura et al.,Am J Respir Cell Mol BIol,1997)。実験的移植血管におけるendothelin受容体の変化を明らかにした(Eguchi et al.,Cardiovas Res,1997)。さらに、rho kinase mRNAの発現について報告を行った(Niiro et al.,Biochem BIophys Res Commun、1997)。また、カルシウム情報系に関し、細胞内カルシウム濃度と張力の同時測定の結果を報告した。
