Research Abstract	副腎白質ジストロフィー(ALD)はペルオキシソーム病の中で最も頻度の高い進行性の脱髄性疾患で、ALD proteinが病因蛋白質であるが、極長鎖脂肪酸活性化酵素(VLACS)の障害が病態と深く結びついていると考えられている。我々はVLACSの精製・クローニングと患者解析を通じてALDの病態生理を明らかにする目的で以下の研究を行った。 (1)VLACSの精製:ラット肝臓のペルオキシソーム画分より精製したVLACSのnative enzymeの分子量は約235kDa,サブユニットの分子量は約70kDaで、極長鎖の脂肪酸基質に活性を有し、既知の長鎖脂肪酸活性化酵素とは異なっていた。 (2)ラットVLACS cDNAのクローニング:ラットVLACSの部分アミノ酸配列よりプローブを作成し、λgtll cDNAライブラリーを用いてラットVLACS cDNAをクローニングした。全長は2972bpで620アミノ酸をコードし、推定分子量は70692で、精製酵素とよく一致した。mRNAは約3kbと推定され、主として肝臓と腎臓で発現しており、ペルオキシソームとミクロソームに局在すると考えられた。得られたcDNAをCOS-1細胞で発現させたところ、約50倍のVLACS活性の上昇を認めた。ホモロジー解析では、fatty acid transport proteinと高い相同性を有していたが、ALDPとは相同性を認めなかった。 (3)ヒトVLACS cDNAのクローニングと遺伝子座位:ラットcDNAをプローブとしてヒトVLACS cDNAをクローニングした。ラットと同じく620アミノ酸をコードしており、得られた最長クローンは2362bpであった。ヒトVLACS遺伝子座位はFISH法により15q21.2と決定された。
