| Research Abstract |
正常ヒト毛包の外毛根鞘にはDOPA陰性の非活性型メラノサイト(ORS-MC)が存在し、この細胞はpre-melanosome関連抗原を有するがmelanosome関連抗原やmelanin合成酵素であるtyrosinase,TRP-1,TRP-2を欠くことを示した(J.Invest.Dermatol.106:28,1996)。また本細胞はTPA添加培養液中で培養でき、その場合はDOPA陽性となることを報告した(J.Derm.Sci.12:204,1996)。またmelanocyteのTPA非添加培養液を用いた培養法について報告した(Pigment Cell Res.9:58,1996)。またmelanoma細胞がPKCαまたはβを欠いておりTPAにより増殖抑制が起こることを示した(J.Cell.Physiol.167:406,1996)。一方、melanomaやmelanocyteの表皮内での重要な接着分子の一つであるE cadherinの表現について検討し、melanoma progressionやUVB照射後E cadherinは減少することを報告した(J.Invest.Dermatol.106:1320,1996)。これらのことよりORS-MCは紫外線照射によって活性化を受けDOPA陽性となりE cadherinなどの接着分子の発現が減少し遊走しやすくなるという可能性が示唆された。また抜毛した毛包よりmelanocyteが培養できることを示したことは今後のmelanocyte biologyの研究には有意義であり現在さらにORS-MCのin vitroでの解析を継続中である。またこれらの基礎研究をふまえてpiebaldism(皮膚38:342,1996)および尋常性白斑(Abstract,Pigment Cell Conference in Anaheim,1996)の表皮移植後の色素拡大現象について報告した。
