Research Abstract |
C57Bl/6J mice were anesthetized by nembutal and treated with thoracic irradiation to dose in the range 15 to 30 Gy. In mice treated with 25 and 30 Gy body weight losses were manifest at 1 to 12 months after irradiation. Deaths (22%-40%) were observed in the period 180 days after 30 Gy irradiation. Delayed administration of exogenous human recombinant hepatocyte growth factor (hrHGF, 4μg/mouse/4 days, I.v.) was performed with mice at 10 days after irradiation. All mice treated with hrHGF were lived for 1 year after 30 Gy irradiation. C3H/HeN mice were treated with and without hrHGF injection (6μg/mouse/6 days,) at 11 days after 15 Gy irradiation. Mice both groups were alived for 240 days. Improvement of acute pneumonic and fibrotic changes by hrHGF injection was unclear based on the histological observation. Relative content of collagen to total protein in the lung at 10 months after irradiation was not different among four treatment groups: 0 Gy, 15 Gy, 30 Gy,+HGF. HGF might be give protection of the lung during the early pneumonic phase after irradiation. Concentrations of endogenous HGF in liver, kidney and lung in ICR mice exposed to radiation depended on the dose and the kind of organs. HGF contents in both liver and kidney after 30 Gy total body irradiation (TBI) increased at 12 hrs temporarily, lung reached to maximum at 48 hrs. after 2 Gy TBI, only lung showed the increase of HGF at 72 hrs. HGF may act as a factor for repair of radiation induced injury in the lung.
