Research Abstract	12および24カ月齢のC57BL/6J雌マウスの末梢血、脾臓、骨髄の構成細胞の異常の有無を検討するため、ガラスホモジナイザーを利用して遊離リンパ球を集め、モノクローナル抗体L3T4、B220、Mac-1により、単色膜免疫蛍光抗体染色法を行い、FACScanで解析を行った。 1.末梢血:24カ月齢のマウスでは12カ月齢よりB220陽性細胞の割合が増加し、逆にMac-1陽性細胞の割合が減少した。L3T4陽性細胞については有意な増減は認められなかった。 2.脾臓:末梢血同様、24カ月齢のマウスでは12カ月齢よりB220陽性細胞の割合は増加したが、Mac-1陽性細胞の割合は小リンパで減少、大リンパ球で増加傾向がみられた。L3T4陽性細胞については明らかな増減は認められなかった。 3.骨髄:24カ月齢のマウスでは12カ月齢よりMac-1陽性細胞の割合の減少傾向があり、B220陽性細胞およびL3T4陽性細胞については明らかな増減は認められなかった。加齢に伴って惹起されるアミロイドーシスの発症機転には何らかの免疫異常の関与を考えている。現在この意義について検討中であり、さらに若年マウスと比較する予定である。また現在、種々の細胞外マトリックスのプローブを用いて腎臓の、SAAやSAPのプローブを用いて肝臓のノーザンブロットを行い、アミロイドーシスの発症機序に関わる物質を検索中である。
