Research Abstract

By using variety of condition of electroporation technique, Lucifer Yellow was induced into YTS-1 (human transitional cell carcinoma cell line). The best efficacy, was obtained with using ilectric field of "100 micro second squar-wave, 1Hz 8 pulses, 1000-1750 V/cm. Under the condition, the induction rate was 70-80%.
Using this condition, Bleomicin, Adrinamycin or Cisplatin was induced into YTS-1. Two hours after the procedure, intracellural drug concentrations were estimated. Compared with the non-electroporated cells, electroporated cells showed 1.5-2.5 fold higher drug concentrations (P<0.01). Citotoxicity of each drug was estimated by ^3H-thymidine uptake assay. Cytotocxicity of Adriamycin and Cisplatin showed same cytotoxicity to the electroporated cells as 10^1-10^2 fold concentrated drugs to non-electroporated cells. Moreover, Bleomycin showed same cytotoxicity to the electroporated cells as 10^3 fold concentrated drugs to non-electroporated cells. Thus Bleomycin was considered to be the most suitable drug to the electroporation technique.
Rats bladder tumors were induced by BBN and the effect of Bleomycin with or without electroporation was estimated histologically. In the Bleomycin plus electroporation group, damages were observed histologically in 60-66% of tomers. On the other hand, no damage was seen in Bleomycin alone group, electroporation alone group and no therapy group.
Thus, the usefullness of electroporation with Bleomycin was proven.
