妊娠初期正常トロホブラストと腫瘍性トロホブラストの増殖・浸潤能の差異の検討

| Research Abstract |
妊娠初期のトロホブラストは増殖能が高く,異種組織である母体の子宮内膜に侵入する現象は癌細胞の増殖・浸潤と類似するが,トロホブラストの増殖・浸潤は,厳密にコントロールされており,なんらかの調節機構が存在することが考えられる。われわれは,妊娠初期の各種トロホブラストにおいて,細胞周期関連因子・アポトーシス・アポトーシス関連分子がどのように発現しているかを組織化学的に検討し,さらに脱落膜内に浸潤する中間型トロホブラスト(intermediate trophoblast)と,その腫瘍化したplacental site trophoblastic tumor (PSTT)で増殖能の差異を検討した。その結果,妊娠初期絨毛のトロホブラストの中では,trophoblasts in the cell columns (CC)とcytotrophoblast (CT)の増殖能が高く,多様な細胞周期関連因子の発現も認められた。逆にsyncytiotrophoblast (ST)とintermediate trophoblast (IT)の増殖能は低く,細胞周期関連因子の発現はあまり認められなかった。また,placental site trophoblastic tumor (PSTT)は,正常のITよりも明らかに増殖能が高く,細胞周期関連因子の発現にも異常を認め,PSTTでは,正常のITで存在する細胞増殖調節機構に破綻をきたしていることが示唆された。STでは,散在性にアポトーシス細胞を認めたことから,STの増殖抑制にアポトーシスが関与している可能性が示された。さらに,STではbcl-2の強い発現を認めたにも関わらず,アポトーシスが起こっていることから,STにおけるアポトーシスの誘導に,bcl-2を介さない系の存在することが示唆された。
