Research Abstract |
Thirty six women, possessing of sperm immobilizing antibody in their sera were examined of their HLA-DR and DQ types using DNA obtained from peripheral blood white cells. The typing of HLA-DR and DQ locus were performed by PCR-SSO,PCR-SSOP and PCR-RFLP.The determined haplotype frequencies of patients were estimated of statistical significance, compared to normal Japanese populations (986 haplotype for HLA-DRB1,1068 for HLA-DQA1 and 2432 for HLA-DQB1). There were significant increase in phenotypes of HLA-DRB1 and HLA-DQB1 locus but not in HLA-DQA1 locus. There were 25 gene frequency percent for HLA-DRB1**0901 in contrast to 13.6 percent in normal population (p<0.0085) , 12.5 percent of patients of HLA-DRB1**1501 in contrast to 6.7 percent of normal population (p<0.0025) , 13.9 percent in contrast to 6.8 percent of HLA-DQB1**0602 (p<0.0089) and 25 percent of patients in contrast to 14.8 percent of normal population of HLA-DQB1**0303 (p<0.0018). The most strong disequilibrium found in HLA-DRB1**0901-DQB1**0303 haplotype among infertile women possessing sperm immobilizing antibody. This haplotype of HLA-DRB1**0901-DQB1**0303 is very rare in Caucasians (from 0 to several percent of haplotype frequency) but characteristic high in Japanese (about 13 percent of that). The high frequency of HLA-DRB1**0901-DQB1**0303 among the patients may account for higher frequency of sperm immobilizing antibody in Japan compared with another country reports. It was elucidated that anti-sperm immunity of human was restricted and controlled by HLA-DR or DQ genes.
