Research Abstract |
Alloplasts have been used to facilitate the regeneration of the periodontal regeneration suffered from periodontal disease. We have developed bioabsorbable ceramics, which was called biphasic calcium phosphate (BCP) granules, for clinical use. Recently, there have been attempted to apply recombinant human bone morphogenetic proteins (rhBMPs) in medical and dental treatments. The purpose of this study was to investigate the ectopic bone formation induced by BCP granules combined with rhBMP-2 in rat dorsum. 1) Ectopic bone formation by BCP combined with rhBMP-2 in the rat dorsum These results indicated that BCP combined with rhBMP-2 induced ectopic bone formation without additional carriers. Therefore, BCP granules alone can function as carriers for rhBMP-2 to induce bone formation. 2) Ectopic bone formation by three types of ceramics combined with rhBMP-2 in the rat dorsum Biphasic Calcium Phosphate (BCP), beta-tricalcium phosphate (TCP) and hydroxyapatite (HAP) were used as carriers for rhBMP-2. These results showed that new bone formation was significantly greater in the BCP-BMP than in the TCP-BMP and the HAP-BMP. 3) Longitudinal observation (four days, one weeks, three weeks, twelve weeks postoperation) of ectopic bone formatin by BCP combined with rhBMP-2 in the rat dorsum A large quantity of new bone was formed from 4 weeks after operation. Twelve weeks postsurgery, a large quantity of new bone has resided without resorption. Consequently, BCP granules alone would be applicable as a carrier for rhBMP-2 in order to induce new bone formation.
