Research Abstract |
Histoculture drug response assay was performed in the 19 patients with advanced oral squamous cell carcinoma (23 specimens) at the department of oral surgery, and its usefulness was assessed. In this assay, drugs dorrved including cisplatin, carboplatin and adriamycin were used. Specimens from a primary carcinoma biopsy, and carcinoma tissue specimens during operation surgical were cultured in contact with the drugs on collagen gel for 7 days. More than 50% carcinoma-inhibiting effects by the drugs in MTT assay to the control was considered useful. Drugs were cut off at the concentration of 20 mug/ml of CDDP, 25 mug/ml of CBDCA, and 15 mug/ml of ADM.This histoculture drug response assay could keep inter cellular contact during culture. The effect of the drugs with MTT assay was assessable in 91.3% of cases of oral squamous cell carcinoma, as confirmed with MTT assay. Primary region, metastasis lymph node and each parts had no different inhibition index. Althrough there was no significant differnce between chemotherapy and non-chemotherapy based on CDDP or CBDCA in 10 assesable cases, this could be one of the could be one of anticancer drug choises. This method appears to be useful as a clinical test.
