Research Abstract |
To study the role of stress protein in homeostatic regulation of biological systems as a molecular chaperone, we investigated the effect of aging, dietary restriction and chronic ingestion of alcohol on the expression of Hsp70 in rat. The amount of Hsp70 was measured in brain and liver of non-stressed 5-120 week-old Wistar rats. Hsp70 was composed of much amount of constitutively expressed form and a little amount of heat-inducible form. A group of rats was restricted their food every other day for 14-38 weeks. Caloric restriction is the only experimental manipulation known to retard aging and increase survival of mammals. Another group of rats was treated with ethanol consumed a vitamin/mineral-fortified liquid diet containing 5% (v/v) ethanol for 6 weeks. The amount of Hsp70 was detected with anti-Hsp70 antibody. (1) It was found that the expression of Hsp70 in brain increased with aging, while not changed in liver. It was suggested that Hsp70 has a significant role in a post-mitotic cell. (2) The amount of Hsp70 in each area of brain, e.g., hippocampus, striatum, thalamus, cerebral cortex, was a little different from each other. (3) The amounts of Hsp70 in brain was significantly lower in rats dietary restricted than those fed ad libitum. It might be caused by decreased stress such as oxidative stress. (4) The chronic ingestion of alcohol affected on the expression of Hsp70 in aged rats, while not changed in young rats. It was indicated that the expression of Hsp70 in brain was altered with aging and modified by dietary restriction and chronic ingestion of alcohol. These results suggest that the altered expression of Hsp70 in brain was caused by the altered rate of stress with aging and stress protein participated in aging of brain.
