Research Abstract |
We have found that peptide growth factors such as VEGF suppress the apoptotic cell death of the hematopoietic cells induced by exposure to gamma-rays. 1. To elucidate the molecular mechanisns of the suppressive effects of VEGF,we examined the mRNA level and the protein level of the bel family (BCL-2, BCL-XL,MCL-1) by Northern blot and Western blot hybridization analysis. The mRNA level and the protein level of MCL-1 were increased in the CMK86 cells, a human leukemia cell line, stimulated by VEGF.Those of BCL-2 or BCL-XL,however, did not change by VEGF.MCL-1 may be involved in the suppressive effect of VEGF on apoptotic cell death. 2. We cloned a novel Kruppel-type zinc finger gene ZK-7, induced by VEGF from a CMK86 cDNA library. The deduced ZK-7 protein has 298 amino acids, and has KRAB (Kruppel associated box) -A box at the N-terminus and seven C_2H_2-type zinc-finger domain at the C-terminus. The function of the ZK-7 gene on the signaling pathway of popsis remains to be elucidated.
