Research Abstract	糖蛋白質と共に細胞表面の「顔」としての様々な働きを担っている酸性糖脂質ガングリオシドは、糖鎖の多様性を通じて分子認識、細胞の分化・発生,免疫機能の発現等に関与していることが多数の生理学的・免疫学的研究により示されてきた。現在の課題は、細胞間認識における直接的関与、細胞と基質の接着の調節機能、細胞膜情報伝達調節機能等の機構解明であり、細胞のガン化、ガンの浸潤、転移等の極めて重要な課題を解決する鍵を握っていると考えられている。しかし、残念ながら、構造物性的研究では多量の試料を要するため、ガングリオシド分子の凝集系の構造、構造安定性・可変性、蛋白質との相互作用等に関する構造物性学的、物理科学的な知見は極めて乏しい状況にあった。そこで、本研究課題では、構造物性的な観点からガングリオシドの機能構造を明らかにする目的で、放射光X線及び中性子線を用いてガングリオシド分散系の溶媒条件をパラメーターとして構造安定性に関する研究を実施し、実験散乱曲線から凝集体の内部構造と表面電荷を直接評価可能な新たな解析法等を開発して、温度変化に伴う糖脂質頭部領域の収縮と熱履歴、有効電荷の変化(ガングリオシドモノマーの解離度の変化)、糖鎖頭部領域の熱可逆的な多量の水の吐出・吸蔵等の極めて興味深い知見を得た。これらの結果は、細胞膜表面に存在するガングリオシド自身が外部環境の変化にダイナミックに応答し、生体膜の界面構造や表面電荷分布に局部的な変動や揺らぎを生じさせることを示唆し、また、蛋白質や高分子の膜界面への吸着、境界脂質で安定化された膜蛋白質の膜内充填状態や構造、膜蛋白質間相互作用などの変化を誘導する可能性も示唆している。また、細胞膜表面おける糖鎖による分子認識モデル系の研究(ガングリオシドミセルとモデル蛋白質との相互作用)を実施した結果、非特異的相互作用ながらモデル蛋白質表面とガングリオシド糖鎖の組み合わせの違いにより、複合体形成過程が著しく異なることなどを見い出した。
