Research Abstract

We investigated in vitro and in vivo osteogenic ability of cultured marrow cells by gene expression analysis of alkaline phosphatase (ALP), osteocalcin (OC) and c-SRC.In the initial stage of bone formation by cultured marrow cells, ALP, OC and c-SRC mRNA could be detected in vivo and in vitro. These are useful as a marker of bone formation to obtain valuable information about osteogenic response of cells on biomaterials. We succeeded in production of cultured bone/ceramic constructs which has high osteogenic potential in vivo. A significant benefit of this method is that the construct can be made with only a small amount of aspirated marrow cells from aged patients with little host morbidity. The analyses of these genes showed that we can assemble an in vitro bone/ceramic construct, and due to the porous framework of the ceramic, the construct has osteogenic potential similar to that of autologous cancellous bone.
