低酸素環境下における口腔内好中球の代謝動態

Research Abstract

ヒト口腔内白血球の簡便な単離法を確立し、その活性酸素の産生の量と質を解析し、口腔内白血球が刺激物非依存性に活性酸素を産生していること、さらにその活性酸素分子種はOCL-が主体であることを明かにした。また、末梢血好中球が腫瘍壊死因子(TNF-α)や顆粒球コロニー刺激因子(G-CSF)などによりprimingされた際に並行して起こるチロシンリン酸化が未刺激の口腔内好中球にも検出され、自発性の活性酸素産生がgenisteinやherbimycin A などのチロシンキナーゼ阻害剤で抑制されるが、H-7やcalphostin C などのC-キナーゼ阻害剤では抑制されないことを明らかにした。さらに活性化に関わるチロシンリン酸化酵素がsrcファミリーのlynおよびSykであることを明らかとした。また、細胞を低酸素下に培養するとスーパーオキシド産生は低下し、NO産生が主になることが判明した。さらに基質となる115kDa蛋白質がc-Cblであることも明らかにした。これにより本細胞が口腔内で常に活性酸素を産生することにより、生体防御作用を発現していることが示唆された。さらに、口腔内好中球からは無刺激で一酸化窒素(NO)産生が認められた。解析の結果、この産生には誘導型NO産生酵素(iNOS)の発現によることが明らかとなった。また、口腔内好中球は、口腔滲出時に口腔内に活性酵素の代謝制御物質などが存在すると影響を受けるが、これが口腔内の防御系にどの様な意義を有するかを解析した結果、お茶に含まれるポリフェノール類のカテキンやタ-メリックに含まれるクルクミンは、自発性活性酸素産生を低濃度で抑制することが明らかとなった。しかも、本抑制作用は、チロシンリン酸化酵素に依存しており、これまで報告されているラジカルスカベンジ作用やC-キナーゼの作用は弱いことが明らかとなった。口腔内好中球は末梢血好中球に比べて短時間でDNA fragmentationを起こすことが示された。大気下では口腔内好中球は末梢血好中球に比べて短時間でDNA fragmentationを起こすが、低酸素環境下ではその寿命が延長することが判明した。本細胞死にも酸化ストレスが関与していることが判明し、アスコルビン酸やグルタチオンやシステインといったSH化合物の添加により細胞死が抑制されることが判明した。
