| Research Abstract |
本研究の主な研究成果を以下に列挙する。(i)既に構造や脳内分布の明らかにされた、オピオイド受容体相同物(ROR-C,MOR-C)の内在性リガンドの同定を行った(Mol.Brain Res.in press)。実際にはスイスやアメリカの研究グループに先んじられたが、私も独立に脳内に存在するペプチド性のROR-Cリガンドを同定した。オピオイドリガンドとは逆に発痛因子として機能するとのデータから、その新規ペプチドはノシセプチンと命名され、ROR-Cオピオイド受容体相同物はノシセプチン受容体と呼ばれることになった。(ii)ノシセプチンは他の神経ペプチドと同様に前駆体から切り出され生成する。私はそのノシセプチン前駆体の全一次構造と脳内分布をそのcDNAクローニングから明らかにした(Biochem,Biophys.Res.Commun,219,714-719)。(iii)私はノシセプチン受容体欠損マウスの作製と解析を行い、その生理的機能を明らかにした(EMBOJ.in press)。得られた変異マウスはノシセプチン投与による疼痛閾値の低下や自発運動量の減少は欠失しているが、正常時の疼痛知覚や運動量には異常は見られない。これらの研究により、ノシセプチン、ノシセプチン受容体系の中枢神経系での生理的存在意義をほぼ明らかにすることができた。しかしながら、実際のノシセプチン-ノシセプチン受容体系の調節部位の同定など不明な点も多く残されている。
