Research Abstract |
1. The research of natural medicinal resources for search of anticancer substances in Madagascar : The research areas with traditional herbal medicines and with unused plants were limited to Fort-Dauphin, Ambosary and Morondava district in Madagascar. The 40 plant species were collected in these areas during out field works. We tried to screening by anticancer promoting activity for 3 species selected from among the collection plants. From the results of this test on each methanol extract of the 3 plants, two kind plants, Hypoxis angustifolia (root) and RESONDRO (vernacular name) showed anticancer promotion activity and these data were as followed ; the former control % and (viability %) were 30.8 (70) on 10μ/ml (conc., ) and 70.2 (70) on 1μ/ml. e structures of six new compounds and six known compounds from Uvaria acuminata, which were dihydrochalcons and acetogenins. Giliama people of southeast region in Kenya use this plant for traditional medicine of painkiller or antibacterial agent. Two dihydrochalcons, 5''''-(2''''-hydroxybenzyl)-diuvaretin and 5'''-(2''''-hydroxybenzyl)-diuvaretin were afforded for anticancer screening test using HCC-Panel (human cell line panel). Then, the data computerized from the HCC-panel screening system showed a unique Mean Graph (Finger Prints), and it was suggested that the compound possess a unique anticancer active mechanism. On the other hand, these anticancer active dihydroxybenzly chalcons in HCC-panel system inhibited Topo-I and -II isomelase action. The comparison between the HCC-panel activity and Topoisomelase inhibition of a same compound is of interest from the viewpoint of correlation between chemical structure and bioactive mechanism of these compounds. Further, we isolated a new pyrene compound from Uvaria lucida of another species of genus Uvaria. The structure of this compound was particularly elucidated by spectroscopic analysis. The compound appeared inhibitory effect to cancer promotion on Epstein-Barr Virus (EBV). The inhibitory effect the pyrene compound on EBV activation ; control % (viability %) =50.9 (60) on 10μg/ml.
