Research Abstract	循環系の恒常性維持機構をよりよく理解するために,比較分子生物学的なアプローチを試み,以下の成果を得た。(1)ウナギで見つけた新型受容体の機能解明:私たちが見つけた新型受容体(D型)はRNase Protection Assayの結果から脳に多いことがわかっていたので,さらに脳のどの部分に多いかを抗体染色で調べ,視索上核のバソトシンニューロンと小脳のプルキンエ細胞に多く発現していることを明らかにした。(2)海水適応に重要なKチャンネルの発見と性質の解明:ウナギを淡水から海水に移したときに誘導されてくる分子の1つとしてKチャネルを同定し,それが新型の内向き整流性Kチャネルであること明らかにした。全一次構造も決定した。ついで,哺乳類のホモログの探索を試み,ラットでは苦戦をしいられているが,ヒトでは成功しそうなところまでこぎつけている。(3)ウナギのA型ナトリウム利尿ペプチド受容体(NPR-A)と海水適応:これまで未クローニングであったウナギのNPR-Aをクローニングし構造を決めた。NPR-Aが海水適応時のNaの排泄に重要な働きをしていることも生理学的な測定から明らかにした。(4)先祖型としてのCNPから受容体の先祖型を探る:サメにはCNPのみしか存在せず,従って,CNPがナトリウム利尿ペプチドファミリーの祖先型であることを示した。従って,サメの受容体を解析すれば,受容体(NPR)の祖先型に関する情報が得られることになる。そこで,まずリガンド結合実験により受容体の性質を明らかにして,現在受容体のcDNAのクローニングを行っている。(5)酸性適応機構解明の糸口:酸性適応能を有するウグイを材料に,Differential Displayにより候補分子を数種類単離した。
