組み換え近交系マウスを用いた肺腫瘍抵抗遺伝子の研究

Research Abstract	肺腫瘍高感受性のA/Jと低感受性のSM/J系の間で26系統からなる組み換え近交系SMXAを樹立し、ウレタン誘発肺腫瘍発生率を調べたところ、その1系SMXA24は既知の4つの感受性遺伝子が感受性アレルをもつにも関わらず、強い抵抗性を示した。A/Jへの戻し交配系にウレタンを投与し、発生した肺腫瘍数を定量的形質バラメターとして遺伝解析を行った結果、第11、第12染色体上にそれぞれ有意のQTLを検出した。これらはいずれもSM/J由来アレルが優性で肺腫瘍敷を抑制した。染色体位置から第11染色体上のQTLはDraganiが報告したPar1と同じとみなされたが、第12染色体上のQTLは未報告の抵抗遺伝子でPar3と命名された。マップ位置からPar3の侯補遺伝子はnPKCηである。nPKCηは肺、皮膚で特異的に発現され、肺腫瘍で抑制されることがわかっているが、これまでの検索ではその発現、PCR産物の構造等についてみるべき異常はみつかっていない。ヒトの肺腺癌材料薬50例についてPar1,Par3の相同染色体部位のLOHを検索したところ、informative caseのうち、前者で50%、後者で30%の頻度でLOHが検出された。比較遺伝学的はヒト固形癌の遺伝的異常を検索するひとつのアプローチである。
