Research Abstract	本研究は、ヒト肝癌発症の原因ウイルスと考えられるB型およびC型肝炎ウイルスの機能的受容体の構造と性質を解明し、ウイルス性肝癌の発症機構における宿主受容体の役割を明らかにする。これにより肝癌発症の予防、診断、治療への基礎的情報を得ることを目的としている。本年度ほ成果として下記の結果が得られた。 1)B型肝炎ウイルス表面抗原(HBsAg)粒子に結合性を示す微量糖タンパク質をヒト肝臓から部分精製することが出来た。 2)この糖タンパク質上のシアル酸含有糖鎖が、受容体活性に重要であり、Neu5Ac α 2-6Gal、Neu5Ac α 2-3Galおよび、Gal β 1-3GalNAcを含有すること、O-シアログリコプロテインエンドペプチダーゼ処理によりムチン型(O-グリコシド型)糖鎖を含むこと、N-グリコシド型糖鎖も存在することなどを見い出した。 3)さらに、この糖タンパク質に対する単クローン抗体を作製することに成功した。得られた単クローン抗体のサブクラスは、IgG1kであり、本糖タンパク質の糖鎖を認識しているものも得ることが出来た。この抗体は、HBsAg粒子感染機構の解明に有用であることが期待された。以上、本研究により、HBVの機能的受容体の構造の基本的性質が明らかにされたが、その機能、生体における分布、遺伝子、さらに感染における役割、発癌への移行メカニズムなどはこれからの研究に待たなければならない。さらに本研究を継続できればと考えている。
