| Research Abstract |
ヒトのがんの90%は上皮細胞に由来するがん腫(carcinoma)である。本研究は上皮組織の形態形成、増殖、分化などの分析を通じて、ヒトがんの発生と進展のメカニズムを明らかにすることを目的に設定された。具体的には次の項目を明らかにする。対象とする上皮組織は、表皮ケラチノサイト、子宮頚部扁平上皮、肺胞上皮、胃上皮などである。
1. 上皮系細胞の発生と形態形成:重層扁平上皮系における幹細胞候補を、p75NGFR,Integrinなどの分子の発現により同定した。候補細胞は培養条件下で増殖能を示した。皮膚の発生において胎児期に発現する遺伝子をDifferential RNA Display法によって数種クローニングした。
2. 上皮細胞系の増殖制御:胃粘膜のパラ分泌増殖因子TRを分離、その作用機序を明らかにした。子宮頸部扁平上皮を用い、上皮細胞の増殖停止に高発現する遺伝子を分離した。
3. 上皮細胞系の分化:扁平上皮細胞系の終末分化において、分化の誘導とG1期停止がそれぞれ異なるメカニズムによってシグナル伝達されることを見いだした。
