| Research Abstract |
EBV DNAのBamHI-X断片を含むXbaI断片12.3kbpのSmaI部位へHIVのenv、rev及びネオマイシン抵抗性遺伝子(全体で6.3kbp)を挿入したプラスミドを作製した。得られたプラスミドをAkata細胞(細胞当たり約20コピーのEBVプラスミドを含む)へ電気穿孔法により導入した。G418を700mg/ml加えて培養を行い、G418抵抗性の細胞クローンを分離した。Southern blot法により相同組み換えの起こった細胞クローンを同定した。
得られた細胞に抗ヒト免疫グロブリン(Ig)抗体を加え、EBウイルス産生を誘導し、24時間後に培養上清を回収した。この上清中には組み換えEBVと共に野生株EBVが含まれる。この混合EBV液をEBV陰性Akata細胞へ感染し、薬剤選択により組み換えEBVのみが感染した細胞クローンを分離した。調べたクローンすべてで組み換えEBVのみが感染していた。
組み換えEBVを末梢血リンパ球に感染し、不死化リンパ球を得た。envの発現がWestern blot法により確認された。
今後得られた組み換えEBVを用いてHIV特異的CTLを誘導する予定である。
