| Research Abstract |
1)bFGFと共同で血管増殖を示す合成ピリミジン化合物を投与することにより、マウスにおける排卵数が2倍に増加した。排卵した卵子は正常に受精し、胚盤胞へ発育した。
2)卵母細胞のGV期には不活性型MAPキナーゼが細胞質にあり、GVBD前に一部のMAPキナーゼは活性化し、GV内に移動することを明らかにした。また外因性の活性型MAPキナーゼをGVに導入すると、GVBDが誘起される。一方、卵子卵丘複合体で合成されるグリコサミノグリカンの動態を解析し、a)卵胞刺激ホルモンや卵胞液にグリコサミノグリカン産生促進作用がある、b)産生されるグリコサミノグリカンはヒアルロン酸とコンドロイチン硫酸であるが、量的にはヒアルロン酸が圧倒的に多い、c)卵子がヒアルロン酸の産生を促進する、d)コンドロイチン硫酸の産生に卵子は影響しないことを明らかにした。インテグリンが精子受容体として機能することやリン酸により誘導される2細胞期発生停止はG2/Mでの停止であり、ras、rafを介してMAPキナーゼが機能することを明らかにした。
3)卵母細胞にFas、顆粒膜細胞にFasリガンドの発現を認めた。またFas-Fasリガンドシステムにより誘導されるDNAフラグメンテーションは透明帯によりブロックされた。
