| Research Abstract |
細胞周期時期特異的に分解されるサイクリンは、その蛋白分解と細胞の生理機能との関係が具体的に解析されている蛋白質である。本研究では、細胞周期の調節機能を担うサイクリン蛋白質の機能ドメインを解析するため、サイクリンの分解機構とM期終了におけるサイクリン分解の調節的役割に焦点を絞り、in vivo及びin vitroでのサイクリン分解の機能ドメインの解析を行なった。卵抽出液によるサイクリン分解の機能ドメインのin vitro解析とサイクリンの酵母への導入によるin vivo解析を通して、サイクリンA,サイクリンBの分解に関与する機能ドメインを解析した結果、サイクリン分解に必要な領域が、M期サイクリンのN末端領域の分解ボックス及び分解ボックスの下流側N末端ヘリクッスに存在することがわかった。次に、分解ボックスとcdk結合部位サイクリンボックス間の保存領域であるN末端ヘリクッスに注目してその機能を解析した結果、N末端ヘリクッスはCdc2と弱い相互作用を介して、サイクリン分解機能に関与することがわかった。次に、XenopusサイクリンA1のN末端ドメインをベイトにした酵母2-ハイブリッドにより、成熟卵母細胞cDNAライブラリーから、サイクリンA1と特異的に相互作用する新規のユビキチン関連遺伝子を同定した。この新規因子は、サイクリンAに保存されたN末端ドメインの特異的モチーフと結合して、サイクリン-CDK複合体に結合することが判明した。
