Research Abstract	本研究は,NCマウスに自然発症するアトピー性皮膚炎の発症メカニズムを解析し,さらにその修復を目指すもので,本年度はコンベンショナルNCマウスに認められる高IgE血症の誘導メカニズムの解明,特に,リンパ球ならびにマスト細胞の関与の解析,産生IgEと皮膚炎発症との関連性を検証,さらに皮膚炎発症とその病態に関与する細菌感染の解析を主たる研究目標とした。 1)IgE産生誘導にはTリンパ球上のCD40リガンドとBリンパ球上CD40結合,さらにIL-4刺激が必要であるが,NCマウスにおいてはCD40とCD40リガンドの発現およびその結合には異常はなく,Bリンパ球のIL-4感受性が著しく高いことが,シグナルトランスダクションレベルからも明らかとなった。さらに,皮膚炎発症マウスでは,CD4陽性細胞と活性化マスト細胞による,Th-2サイトカインであるIL-4,-5,-6などの産生が顕著であることが判明した。 2)高IgE産生が皮膚病発現の原因になりうるかどうかをより直接的に証明するために,NCマウスとIgEレセプター(FcεRIγ鎖)欠損マウスとのバッククロス交配を行った結果,IgEレセプター欠損NCマウスの作製に成功した。現在,皮膚炎の発症との関連性を検討中である。 3)アトピー性皮膚炎発症の誘因あるいは助長因子と推測されている Staphylococcus aureusの感染の有無を調べた結果,皮膚炎部位から採取した全検査試料からStaphylococcus aureusが分離された。従って,この菌による病態への関与が強く示唆された。
