Research Abstract

1) 佐藤は葉緑体DNA結合蛋白質(CND41)が弱酸性条件においてRubisco大サブユニットを部位特異的に切断すること等、CND41のprotease活性を詳細に解明した。さらに、培養細胞、植物体における時期・組織特異的発現、糖による誘導等を明らかとするとともに、ゲノム遺伝子の単離と構造解析を行なった。また、アンチセンス形質転換体を用いてCND41が植物体の形態形成にも重要な役割を果たすことを示唆した。
2) 杉田はタバコ葉緑体RNA結合蛋白質の葉緑体内の存在量、存在形態、結合RNA分子種の特定等を検討し、葉緑体RNA結合蛋白質がストロマ中のRNAの安定性に関与していることを示唆した。また、核コードのファージ型RNAポリメラーゼ(RpoZ)ホモログをN.sylvestrisより単離し、その構造を決定した。
3) 小俣はラン藻の硝酸同化オペロンの転写制御における「基質による正の制御」と「窒素同化産物による負の制御」の相互関係を明らかとした。また、硝酸能動輸送体(NRT)のRドメインとPII蛋白質がそれぞれNRTの活性を抑制すること、およびNRTとRドメインを介する経路以外に炭酸同化の阻害に応答してNRTを制御する機構を見いだした。
4) 長谷はFdを欠失したラン藻にトウモロコシの葉肉細胞特異的FdIと維管束鞘細胞特異的FdIIを導入し、FdIとFdIIの炭素と窒素同化系への電子分配の効率が異なる可能性を明らかとした。一方、光合成器官と非光合成器官における還元力の利用特性を決定する要因として、FdとFd-NADP^+還元酵素の組み合わせが重要であることを明らかとした。
5) 西村はマイクロボディ酵素の発現制御に関わるalternative splicingを解析し、生じた蛋白質の〔末端の移行シグナルの有無により細胞内局在が制御されていることを明らかとした。また、マイクロボデイ酵素cDNAクローニング、その構造解析、これら蛋白質の強制発現形質転換体を用いて、これら蛋白質がマイクロボディの機能転換、さらには植物体機能において果たす役割を明らかとした。
