Research Abstract

1)佐藤は生理的pHにおける葉緑体DNA結合タンパク質(CND41)のプロテアーゼ活性ついて検討を行ない、変性処理したRubiscoがCND41依存的に分解されること、一方、Rubiscoの活性化、あるいは、DNAの添加によりタンパク質分解が抑制されること等を認めた.さらに、形質転換植物体を用いてCND41が老化時のタンパク質分解に関与していることを示唆する結果を得、現在検討を進めている
2)杉田は葉緑体RNA結合蛋白質が種々のストロマRNAに結合し、RNAの安定性に関与していることを示唆するデータを得た。また、葉緑体遺伝子の転写に働くファージ型RNAポリメラーゼと推定されるRpoT-Cを含む3種のRpoTホモログ(RpoT-A〜C)をN.sylvestrisから単離した。
3)小俣はラン藻の炭素欠乏誘導性オペロンのひとつであるcmpABCDオペロンがHCO_3^-能動輸送体をコードしていることを証明し、このオペロンと他の炭素欠乏応答性遺伝子群の活性化には異なる転写因子が関与すること、また、炭素および窒素に応答して制御されるccmKLMNOオペロンには、炭素、窒素それぞれに応答するcis制御領域があることを明らかにした。さらに、PII蛋白質のリン酸化状態の変化とラン藻の硝酸同化の制御の間には直接の関連がないことを証明した。
4)長谷は葉肉細胞特異的なFdIと維管束鞘細胞特異的なFdIIがPSI周辺の電子の分配を制御することを示した。また、Fd-NADP^+還元酵素(FNR)と亜硫酸還元酵素(SiR)を用いて、植物根に存在するNADPH/FNR/Fdの電子伝達カスケードが非光合成器官におけるFd依存性の代謝反応の還元力供給系として働くこと、及びFdとの蛋白質間相互作用の構造特性を明らかにした。
5)西村はhydroxypyruvate reductaseと葉緑体に局在するascorbate peroxidaseがalternative splicingによって2種類の蛋白質として合成されることを明らかにしている。さらに、このalternative splicingが光により制御を受けていることを初めて明らかにするとともに、splicingのトランス因子であるSRタンパク質のcDNAクローニング及びその発現調節を解析した。
