| Research Abstract |
今年度も多様な生物分子モーターの特性の理解にむけた基礎的研究が多く行われた.まず,細胞質性ダイニンの力発生特性を光ピンセット法によって解析した(豊島).その結果,細胞質ダイニンは低濃度(50μM以下)ATPでは連続的な、高濃度(100μM以上)ATPでは非連続的な動きを示し、昨年報告した軸糸ダイニンと同様に2相性の性質を示すことが明らかになった.このような性質はダイニン特有のものと考えられ,興味深い.また,鞭毛ダイニンでは,クラミドモナスからダイニン内腕のc分子種のみを欠失した変異株が単離された(神谷).その運動性の解析から,このダイニンは鞭毛打の波形よりは頻度決定に重要であることがわかった.このような性質はこれまで外腕ダイニンものとされていたので.この発見により外腕,内腕の機能分化に関するこれまでの考えは修正をせまられることになる.植物ミオシンに関しては,現在知られているミオシン中,最も高速の運動を発生するシャジクモミオシンを培養細胞系で発現する実験に着手した.現在,その頭部と尾部に相当する遺伝子をアグロバクターのプラズミドに組み込んで,タバコ培養細胞で発現させる実験が進行中である(山本).そのほか,タバコ培養細胞そのものにおいて,ミオシン2種の存在を同定し,精製することに成功した(新免).これらの情報は今後植物ダイニンの特徴を理解するうえで重要であろう.また,テッポウユリ花粉管ミオシンの運動性はカルシウムイオンによって制御されているが,今回,その制御にカルモジュリンが関与している証拠が得られた(新免).最後に,グルカン合成酵素(GTF)がデキストラン鎖上を滑りながら酵素反応を行う可能性を追求していたが,前回までに,その可能性を強く示唆する結果を得た.今回,酵素とデキストランの両者に蛍光標識を導入した実験系を作成し,この仮説を直接検証する作業に入った(児玉).
