| Research Abstract |
本研究はタバコのプラスチドより調製したin vitro翻訳系を用いて、プラスチド翻訳の分子機構を解明することを目的とする。
(1) UUG開始コドン
クロレラ葉緑体ゲノムのinfA mRNAの第1コドンはUUGである。これが開始コドンとして働くことをin vitro系で確認した。これは葉緑体mRNAで初めて見い出されたUUG開始コドンであるが、効率は極めて低い。
(2) RNAエディティングの影響
タバコrps 14 mRNAのコード領域内の2カ所のCがUに変換する。この変換が翻訳効率に影響するかどうかをかin vitro系で調べたが、変化ないことを示した。
(3) Shine-Dalgarno(SD)配列の効果
タバコ葉緑体のmRNAにはSD配列を含むものがあるが、この効果を変異mRNAとin vitro系で調べたところ、AUGより5〜10ヌクレオチド前方にあれば著しい効果があることを示した。
