| Research Abstract |
本研究はタバコのプラスチドより調製したin vitro翻訳系を用いて,プラスチド翻訳の分子機構を解明することを目的とする。
(1)ジ・シストロニックmRNAの翻訳効率
psbD mRNAは両端がオーバーラップしているジ・シストロニックmRNAである。psbD mRNAはAUGとGUGのコドンを持つが、後方のGUGのみが翻訳開始コドンであることを決定した。また、両シストロンを切り離したモノシストロニックmRNAの翻訳効率は大きな変化がないことを示した。
(2)30種のtRNAの翻訳は可能か
システィンtRNA遺伝子の破壊を行った株を作製中である。
(3)RNAエディティングと翻訳との関係
単離タバコ葉緑体よりin vitroエディティング系の開発に成功した。これを用いて、psbL mRNAのACGからAUGへの変換のシスとトランス因子を同定した。ついで、生成したAUGが翻訳開始コドンとなることを証明した。
