Research Abstract	食道:扁平上皮において末端に糖鎖と結合するレクチン様構造を有する糖蛋白トロンボモジュリン(TM)の発現を検討した.TM発現の異る培養食道がん細胞を親株より分離・樹立して比較したところ,TM発現の少ない方がマトリゲルに対してより浸潤性を示した.また,口腔の扁平上皮でもTM発現が,浸潤・転移巣で減少していた.また,食道癌においてもMUC1抗原の発現が,患者の予後と相関することを見い出した. 胃:胃癌において,糖蛋白抗原の蛋白部分であるアポムチンMUC1,MUC2の発現を検討した.MUC1(+)例はMUC1(-)例に比較して有意に予後が悪く,MUC2(-)例はMUC2(+)例に比較して有意に予後が悪かった.胃癌においてもムチン抗原の発現が予後と相関することが確認された. 胆道系・肝臓・膵臓:この領域における腫瘍には,それぞれの部位で浸潤性増殖を示す群と,膨張性発育を示す群が見られるが,共に予後が著しく異っている.これらでは,前者にMUC1の発現が有意に高く,後者にMUC2の発現が有意に高かった.この発現は免疫組織化学のみならず,in situ hybridizationを行い遺伝子的にも確認した. 大腸:大腸癌の肝転移形成の際の糖鎖の関与を検討する為にStamper-Woodruff Assayを行った.切除されたヒト正常肝臓へIL-1βを注入しE-selectinの発現を導入し,親株細胞より分離された糖鎖抗原シアリル・ルイスX(SLX)の発現の異る大腸癌培養細胞を用いて接着実験を行い,SLXに依存する接着の存在を確認した.次に大腸の前頑病変である腺腫で,異型度の増加と共に腺管周囲の線維成分が減少することを見いだした.更に切除大腸癌を糖鎖合成酵素阻害剤を加えて初期培養することにより,糖鎖と蛋白の分析を行っている.
