Research Abstract

CD14の研究:遊離型CD14発現トランスジェニックマウス(TM)(M14S)を用いてLPS刺激に対し血中TNF-αやIL-6の低下とShwartzman反応の低下、P.o acnes/LPS刺激マウスにおける肝障害阻止、がん転移亢進などを観察した。CD14N末端部のTMを作出した。ヒト臍帯血管培養細胞(HUVEC)、単球およびMφ細胞株THP-1の全ての細胞にTLR1およびTLR4mRNA発現を確認した。TLR2RNA発現は単球およびTHP-1では強いが、HUVECでは殆ど見られなかった。TLR4発現はFACS法でも観察され、LPS刺激で亢進した。LPS刺激によりHUVECのEセレクチンmRNA発現は亢進し、抗TLR4mAbにより抑制された。FACSでも同様な成績を確認した。LPS刺激によりHUVECのIL-6遊離は亢進し、抗TLR4mAbにより抑制された。
ADAMに関する研究:ヒトとマウスADAM17(TACE)に対するmAbを作成し、単球での恒常的発現と活性化好中球における出現を明らかにした。ConAとIL-2刺激T細胞やNK細胞における陽性細胞増加、を認めた。ヒトの特発性間質性肺炎の肺胞壁Mφと好中球、慢性関節リウマチ滑膜下MφにADAM17陽性細胞を認めた。マウスP.acnes注射後の肺や肝の肉芽腫内Mφと好中球に発現を認めた。マウスADAM15遺伝子構造を明らかにした。LPSやサイトカイン刺激による遺伝子発現亢進を観察した。正常胸腺T細胞での発現は陰性であるが、ConAとIL-2刺激後CD4^+CD8^+とCD4^+CD8^-細胞に強く発現を認めた。正常組織では、脳の錐体細胞とPurkinje細胞、骨髄の巨核球、myeloid細胞、erythroid細胞、脾臓の巨核球、単核球、腎集合管上皮細胞、副腎網状帯細胞、心筋細胞などでの発現を観察した。マウスADAM8は常在Mφと好中球で発現し、LPS刺激で増加を観察した。2種の新ADAMcDNAを検出した。89-2-2はzinc-binding siteのコンセンサス配列を持ち、プロテアーゼの可能性が示唆された。
オステオポンチンの研究:ホモザイゴートTMを作成し、現在免疫および炎症反応を解析中。
