Research Abstract |
We established the method of isolation of pulmonary vascular smooth muscle cells to investigate the ion channel of hypertensive pulmonary vascular smooth muscle cell. Cultured smooth muscle cells had changes in phenotype and there might be comtamination of other types of cell such as fibroblast. So, we estimated that isolated pulmonary artery vascular smooth muscle was more suitable than cultured cells. Isolation of pulmonary arterial smooth muscle cells was performed in clcium free Tyrode solutioncontaining 20mg collagenase. The cell suspended solution was passed through the nylon mesh. In chronic hypoxia-induced pulmonary hypertensive pulmonary arteries, KCL-induced depolarization-induced-contractions were reduced by NO doner with small changes in intracellular calcium concentration. NO might after calcium sensitivity of pulmonary arterial smooth muscle cells especially in pulmonary hypertension.
