前立腺癌における転移抑制遺伝子の同定と転移を中心とした癌進展の機序の解析

Research Abstract

前立腺癌の進行に関連した抑制遺伝子を臨床検体からクローニングすることは、その手術症例数が比較的少ないことや癌組織の採取が難しいこと、また癌組織における癌細胞やその組織構築の多様性などにより非常に困難と考えられる。我々は肺に高率に転移するラット前立腺癌細胞にヒト染色体を1本導入し、その肺転移能などの性状の変化を指標としながら転移抑制遺伝子をクローニングする方法を開発してきた。これは目的とする抑制遺伝子を含むヒト染色体小断片を、ラット癌細胞の中に存在させてからクローニングしていくため、臨床検体から同定するよりも効率が良い。
我々はこの方法を用いて、KAI1/CD82遺伝子を11p11.2を含むヒト11番染色体小断片よりクローニングし、この発現がヒト前立腺癌の進行と逆相関していることを示した。ヒト前立腺癌において同領域のLOHを検索したところ、その頻度は低く、この蛋白の発現は他の遺伝子の制御を受けている可能性があるという結論を得た。
またこの方法を用いて8p21-p12にも転移抑制遺伝子が存在することを示し、それ含むヒト染色体8p12小断片(5cM程度)の入ったクローンをすでに得ている。その後1cM程度まで転移抑制遺伝子の存在する領域を限局し、転移抑制遺伝子を含むバクテリア人工染色体を同定し、塩基配列を解析中である。この領域もヒト前立腺癌の進行と関係があり、臨床検体についても解析を進めている。
その他、ヒト7、12番染色体については転移抑制遺伝子が、19番染色体には造腫瘍性抑制遺伝子が存在することを示し、論文として報告した。13番染色体にも転移抑制遺伝子が存在する結果が出ている。ヒト前立腺癌におけるLOHの検索では、13番染色体にLOHが高頻度に認められ論文として報告した。現在7、12番染色体についてLOHを検索中である。
